Retatrutide (LY3437943) is a synthetic triple agonist of the GLP-1, GIP, and glucagon receptors. The GLP-1 and GIP arms work like tirzepatide; the glucagon arm adds a hepatic and thermogenic component, raising basal energy expenditure. Administered subcutaneously once weekly. Investigational — Eli Lilly is currently running phase 3 trials (TRIUMPH program).
